CD(Cluster of differentiation)就是细胞膜表面的分化抗原群/分化抗原簇,应用聚类分析,可将来自不同实验室识别同一分化抗原的单克隆抗体归为一个CD,经由从1982年第1次国际上人类白细胞分化抗原会议(The Human Leucocyte Differentiation Antigen workshop,HLDA) 统一命名。迄今为至,人CD的序号已从CD1命名至CD350 (2009年),可大致划分为T细胞、B细胞、髓细胞、NK细胞、血小板、粘附分子、内皮细胞、细胞因子受体、激活抗原、碳水化合物半抗原、树突状细胞、干细胞/祖细胞、基质细胞和红细胞14个组。白细胞分化抗原(Leucocyte Differentiation Antigen,LDA)是指不同谱系(1ineage)血细胞在正常分化,成熟,活化过程中出现或消失的细胞表面标志,多是跨膜蛋白或糖蛋白,少数为碳水化合物半抗原。随着人类功能基因组计划研究以及免疫学与其他学科的交叉,人类白细胞分化抗原的概念逐渐被人类细胞分化分子(HCDM)所替代。

采用单克隆抗体鉴定方法识别的白细胞分化抗原又称CD (cluster of differentiation) 抗原。 检测CD 抗原是实验室识别细胞及不同分化阶段细胞或细胞亚群最主要的方法。白细胞分化抗原(狭义上的CD)参与机体重要的生理和病理过程:①免疫应答过程中免疫细胞的相互识别,免疫细胞抗原识别、活化、增殖和分化,免疫效应功能的发挥;②造血细胞的分化和造血过程的调控;③参与炎症反应、血栓形成和组织修复;④参与细胞的生长、分化、迁移,及肿瘤的恶化和转移。

CD在基础免疫学研究中主要应用于:①CD抗原的基因克隆,新CD抗原及新配体的发现;②CD抗原结构与功能关系;③细胞激活途径和膜信号的传导;④细胞分化过程中的调控;⑤细胞亚群的功能。CD在临床免疫学研究中主要用于:①机体免疫功能的检测;②白血病、淋巴瘤免疫分型;③免疫毒素用于肿瘤治疗、骨髓移植以及移植排斥反应的防治;④体内免疫调节治疗。

在许多场合下,抗体及其识别的相应抗原都用同一个CD序号,因此在参阅教科书和文献时需加注意:(1)CD是流水编号,CD129暂缺;CD67和CD66b是重复的;CD187-190,CD211,CD237,CD341-343,CD345-248暂未编号。(2)CD中带有w的抗原或抗体CDw12,CDw198,CDw199,CDw293尚需继续进行全面鉴定。(3)有些CD抗原又可进一步划分为不同的成员,一般用小写英文字母表示,但情况有所不同:①如CD1可分为CD1a,CD1b和CD1c,

这三种不同分子是分别由三个不同的高度同源的基因所编码;②CD45至少可分为CD45RA,CD45RB,CD45RC和CD45RO,它们是同一基因的不同异型/同种型(isoform);③CD2和CD2R是识别同一个分子上不同的表位;④CD49已进一步划分为CD49a,D49b,CD49c,CD49d,CD49e和CD49f,它们的基因定位于不同的染色体上,但具有较高的同源性。(4)CD的组划分的特异性是相对的,实际上,许多CD抗原的组织细胞分布较为广泛。此外,有的CD抗原可由不同的分类角度而归入不同的组,如IL-2受体a链CD25是活化T细胞的标记,也属于细胞因子受体;再如某些属于T细胞、B细胞、髓细胞或NK细胞组的CD抗原实际上也是粘附分子,等等。

CD/白细胞分化抗原/细胞膜表面分化抗原群的定义及意义

CD(Cluster of differentiation)就是细胞膜表面的分化抗原群/分化抗原簇,应用聚类分析,可将来自不同实验室识别同一分化抗原的单克隆抗体归为一个CD,经由从1982年第1次国际上人类白细胞分化抗原会议(The Human Leucocyte Differentiation Antigen workshop,HLDA) 统一命名。迄今为至,人CD的序号已从CD1命名至CD350 (2009年),可大致划分为T细胞、B细胞、髓细胞、NK细胞、血小板、粘附分子、内皮细胞、细胞因子受体、激活抗原、碳水化合物半抗原、树突状细胞、干细胞/祖细胞、基质细胞和红细胞14个组。白细胞分化抗原(Leucocyte Differentiation Antigen,LDA)是指不同谱系(1ineage)血细胞在正常分化,成熟,活化过程中出现或消失的细胞表面标志,多是跨膜蛋白或糖蛋白,少数为碳水化合物半抗原。随着人类功能基因组计划研究以及免疫学与其他学科的交叉,人类白细胞分化抗原的概念逐渐被人类细胞分化分子(HCDM)所替代。

采用单克隆抗体鉴定方法识别的白细胞分化抗原又称CD (cluster of differentiation) 抗原。 检测CD 抗原是实验室识别细胞及不同分化阶段细胞或细胞亚群最主要的方法。白细胞分化抗原(狭义上的CD)参与机体重要的生理和病理过程:①免疫应答过程中免疫细胞的相互识别,免疫细胞抗原识别、活化、增殖和分化,免疫效应功能的发挥;②造血细胞的分化和造血过程的调控;③参与炎症反应、血栓形成和组织修复;④参与细胞的生长、分化、迁移,及肿瘤的恶化和转移。

CD在基础免疫学研究中主要应用于:①CD抗原的基因克隆,新CD抗原及新配体的发现;②CD抗原结构与功能关系;③细胞激活途径和膜信号的传导;④细胞分化过程中的调控;⑤细胞亚群的功能。CD在临床免疫学研究中主要用于:①机体免疫功能的检测;②白血病、淋巴瘤免疫分型;③免疫毒素用于肿瘤治疗、骨髓移植以及移植排斥反应的防治;④体内免疫调节治疗。

在许多场合下,抗体及其识别的相应抗原都用同一个CD序号,因此在参阅教科书和文献时需加注意:(1)CD是流水编号,CD129暂缺;CD67和CD66b是重复的;CD187-190,CD211,CD237,CD341-343,CD345-248暂未编号。(2)CD中带有w的抗原或抗体CDw12,CDw198,CDw199,CDw293尚需继续进行全面鉴定。(3)有些CD抗原又可进一步划分为不同的成员,一般用小写英文字母表示,但情况有所不同:①如CD1可分为CD1a,CD1b和CD1c,

这三种不同分子是分别由三个不同的高度同源的基因所编码;②CD45至少可分为CD45RA,CD45RB,CD45RC和CD45RO,它们是同一基因的不同异型/同种型(isoform);③CD2和CD2R是识别同一个分子上不同的表位;④CD49已进一步划分为CD49a,D49b,CD49c,CD49d,CD49e和CD49f,它们的基因定位于不同的染色体上,但具有较高的同源性。(4)CD的组划分的特异性是相对的,实际上,许多CD抗原的组织细胞分布较为广泛。此外,有的CD抗原可由不同的分类角度而归入不同的组,如IL-2受体a链CD25是活化T细胞的标记,也属于细胞因子受体;再如某些属于T细胞、B细胞、髓细胞或NK细胞组的CD抗原实际上也是粘附分子,等等。
